Chloroquine is a 9-aminoquinoline known since 1934. Apartfrom its well-known antimalarial effects, the drug hasinteresting biochemical properties that might be appliedagainst some viral infections. Chloroquine exerts directantiviral effects, inhibiting pH-dependent steps of thereplication of several viruses including members of theflaviviruses, retroviruses, and coronaviruses. Its best-studiedeffects are those against HIV replication, which are beingtested in clinical trials. Moreover, chloroquine has immuno-modulatory effects, suppressing the production/release oftumour necrosis factor -and interleukin 6, which mediate theinflammatory complications of several viral diseases. Wereview the available information on the effects of chloroquineon viral infections, raising the question of whether this olddrug may experience a revival in the clinical management ofviral diseases such as AIDS and severe acute respiratorysyndrome, which afflict mankind in the era of globalisation.
